Tesamorelin is a synthetic peptide that acts as an analog of growth hormone-releasing hormone, a natural hypothalamic signal. Its sequence corresponds to the forty-four amino acid form of the human hormone, with a small acyl group attached near the amino terminus. That modification slows enzymatic breakdown and extends the time the peptide remains active in circulation. The compound was developed as a pharmacological way to raise endogenous growth hormone output rather than supplying the hormone directly.
After injection, the peptide binds receptors on somatotroph cells in the anterior pituitary. Receptor activation raises intracellular cyclic AMP and triggers release of stored growth hormone into the bloodstream. Because the compound works through the body's own regulatory system, growth hormone pulses retain much of their normal feedback control. Repeated administration also raises insulin-like growth factor 1, a hormone produced mainly in the liver. Investigators treat that rise as a marker that the pituitary axis has been engaged.
Tesamorelin binds to growth hormone-releasing hormone receptors on the surface of pituitary somatotroph cells. This binding activates adenylate cyclase, raising intracellular cyclic AMP levels and triggering the release of growth hormone into circulation. The elevated growth hormone then stimulates hepatic production of insulin-like growth factor 1. Because the effect is mediated through the endogenous axis, secretion remains subject to feedback regulation. This distinguishes it from direct growth hormone administration, which bypasses pituitary control entirely.
Clinical investigation has focused on HIV-associated lipodystrophy, a condition in which antiretroviral therapy contributes to abnormal fat distribution. Excess visceral adipose tissue accumulates in the abdomen while peripheral fat may be lost. Tesamorelin was evaluated for reducing this visceral fat depot, with trials measuring changes in abdominal fat by imaging rather than by body weight alone. The rationale rests on the known lipolytic effects of growth hormone. Effects on visceral fat are documented, while long-term outcomes regarding cardiovascular risk remain less clearly established.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, composed of 44 amino acids. It was designed to retain the biological activity of the native hormone while resisting rapid enzymatic degradation. The compound is classified as a growth hormone secretagogue and belongs to the broader family of hypothalamic releasing factors. In research and clinical settings, it is studied for its ability to stimulate pituitary growth hormone release. Its structure includes a modification at the N-terminus that contributes to an extended half-life relative to native growth hormone-releasing hormone.

Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.
Tesamorelin is a synthetic peptide of 44 amino acids that reproduces the sequence of human growth hormone-releasing hormone (GHRH) and carries a trans-3-hexenoyl group on its N-terminal tyrosine. That small fatty-acid modification blocks cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GHRH in plasma. The result is a molecule with a longer circulating half-life than the natural hormone while retaining the same receptor target. It is supplied as a lyophilized powder for reconstitution and belongs to the broader class of GHRH analogs studied for effects on pituitary growth hormone secretion.
Development work on the compound, originally designated TH9507, focused on conditions in which reduced growth hormone signaling is thought to contribute to altered body composition. The United States Food and Drug Administration approved it in 2010 for the treatment of excess visceral abdominal fat in adults with human immunodeficiency virus infection and lipodystrophy. Later research examined other populations, including adults with mild cognitive impairment, where a large trial did not meet its primary endpoints. This mixed record illustrates how a single mechanism can produce clear effects in one setting and inconclusive results in another.
Tesamorelin occupies a narrow position among agents that act on the growth hormone axis. Unlike growth hormone itself, which is given as replacement, it stimulates the pituitary to release the hormone in pulses, so the downstream increase in insulin-like growth factor 1 depends on intact somatotroph function. Other peptides in the same family include shorter GHRH fragments and synthetic secretagogues with different stability profiles. Several points remain unresolved, including whether the reduction in visceral fat translates into fewer cardiovascular events, what happens to metabolic markers after long-term use, and how the drug compares with lifestyle or surgical approaches.
Tesamorelin is a synthetic peptide that belongs to the growth hormone-releasing hormone family and contains the same forty-four amino acid sequence as endogenous GHRH, extended at the amino terminus by a trans-3-hexenoyl group. That small fatty acid modification protects the peptide from rapid cleavage by dipeptidyl peptidase-4, the enzyme that shortens the half-life of native GHRH to only a few minutes. Chemically the compound is produced by solid-phase peptide synthesis, purified by chromatography, and supplied as a sterile lyophilized powder for reconstitution.
Clinical interest in tesamorelin arose from the need to address visceral adiposity in people living with HIV. Antiretroviral therapy improved survival but was associated in some patients with central fat accumulation, altered lipid profiles, and metabolic complications. This condition, often called HIV-associated lipodystrophy, involves excess visceral adipose tissue that is difficult to manage through diet and exercise alone. Investigators evaluated tesamorelin because GHRH analogs can stimulate growth hormone secretion and influence fat distribution without direct liposuction or invasive procedures.
A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
Tesamorelin is a synthetic analog of growth hormone-releasing hormone, a peptide hormone produced by the hypothalamus. The molecule retains the 44-amino-acid sequence of human GHRH and carries a trans-3-hexenoyl modification at its N-terminus. This modification increases resistance to enzymatic degradation and extends the peptide's functional stability relative to native GHRH. The compound is supplied as a lyophilized powder for reconstitution and subcutaneous administration in clinical settings. Its development code was TH9507, and it belongs to the GHRH analog class. It is not a growth hormone product; instead, it acts upstream to stimulate endogenous growth hormone release.
The Badr Organization (Arabic: منظمة بدر Munaẓẓama Badr), previously known as the Badr Brigades or Badr Corps, is an Iraqi Shia Islamist and Khomeinist political party and paramilitary organization headed by Hadi al-Amiri. The Badr Brigade, formed in 1982 and led by Iranian officers, served as the military arm of the Supreme Council for Islamic Revolution in Iraq (SCIRI), a Shia Islamic party based in Iran. The Badr Brigade was created by Iranian intelligence and Shia cleric Mohammad Baqir al-Hakim with the aim of fighting the Ba'athist regime of Saddam Hussein during the Iran–Iraq War. Since the 2003 US-led invasion of Iraq, most of the Badr Brigade fighters have entered the new Iraqi army and police force. Since 2003, the Badr Brigade and SCIRI were considered to be one party, but have recently unofficially separated with the Badr Organization now being an official Iraqi political party. Badr Brigade forces, and their Iranian commanders, have come to prominence in 2014 fighting the Islamic State of Iraq and the Levant (ISIL) in Iraq. It is a part of the Popular Mobilization Forces.
Burger King Holdings was the parent company of Burger King when it went public in 2002. Burger King derived its income from several sources, including property rental and sales through company owned restaurants; however, a substantial portion of its revenue was dependent on franchise fees. During the transitional period after 3G Capital acquired the company, Burger King's board of directors was co-chaired by John W. Chidsey, formerly CEO and chairman of the company, and Alex Behring, managing partner of 3G Capital. By April 2011, the new ownership had completed the restructuring of Burger King's corporate management and Chidsey tendered his resignation, leaving Behring as CEO and chair. Burger King Corporation is currently an independently operated subsidiary of RBI. RBI's present organizational structure includes five primary segments: Tim Hortons, Burger King, Firehouse Subs, Popeyes Louisiana Kitchen and International. International encompasses the aggregated outcomes from the operations of each brand outside the United States and Canada. Josh Kobza, the CEO of RBI, was appointed in 2023. Before taking over as CEO in February 2023, Kobza served as CFO, CTO, and COO of RBI. Tom Curtis, the president of Burger King U.S. and Canada, was appointed in 2021 and oversees the operation of the Burger King Corporation in the United States and Canada. In North America, Burger King Corporation is responsible for licensing operators and administering stores.
== Uterine leiomyomas vs. uterine leiomyosarcomas == Leiomyomas are benign smooth muscle tumors that have overlapping features with leiomyosarcomas. Although both originate from smooth muscle, leiomyomas do not, in contrast, mature into leiomyosarcomas. Leiomyomas are seen in premenopausal women and are symptomatic 20–50% of the time, while leiomyosarcomas, the most common uterine sarcomas, are seen in older postmenopausal women, with 40-60 being the peak age incidence. Since leiomyomas are benign and mostly asymptomatic, minimally invasive treatment modalities are used to treat them. For this same reason, distinguishing them from LMSs before surgical procedures is crucial to ensure that laparoscopic procedures or diagnosis delay will not lead to heightened morbidity given the poor prognosis of LMS. For example, the FDA has warned against using morcellation for benign leiomyomas, as those with unsuspected sarcomas are at risk of cancer spread.
== Diagnosis == Doctors formerly diagnosed cachexia mainly by looking at changes in body weight. A person was considered to have cachexia if they had a low BMI or unwanted weight loss of more than 10%. However, weight alone is not always a reliable method. Factors like fluid buildup (edema), tumor size, and obesity can make it difficult to diagnose cachexia. These weight-based criteria do not account for muscle loss, which is a key part in cachexia. . To improve diagnosis of cachexia, experts proposed adding lab tests and symptom evaluations. With that, a person might have cachexia if they lost at least 5% of their total body weight in 12 months or had a BMI under less 22 kg/m2 with at least three of the following: weak muscles, fatigue, loss of appetite, low muscle mass, or abnormal labs. There have been attempts to define specific types of cachexia, such as cardiac cachexia, which can occur in people with congestive heart failure. However, it has no widely accepted definition.

== Clinical significance == The members of the HtrA family of proteases have been shown playing critical roles in cell physiology and being involved in several pathological processes including cancer and neurodegenerative disease. Strong evidences supported HtrA2's involvement in oncogenesis. This protein is widely expressed in a variety of cancer cell lines, Analysis of biopsy samples showed changes in expression of HtrA2 in cancer tissues compared with normal tissues. HtrA2 has recently been identified as a gene related to Parkinson's disease. Mutations in Htra2 have been found in patients with Parkinson's disease. Additionally, mice lacking HtrA2 have a parkinsonian phenotype. This suggests that HtrA2 is linked to Parkinson's disease progression in humans and mice.
A blue plaque is installed at a branch of Tesco in Walthamstow to mark the October 2022 purchase of the Liz Truss lettuce, which famously outlived her premiership. 6 October – After what The Guardian describes as "months of sniping and criticism" of her, including that her salary was £3,000 higher than that of the prime minister, Sue Gray resigns as Downing Street Chief of Staff. In her resignation statement, Gray says she "risked becoming a distraction". Morgan McSweeney, a person with whom Gray is said to have disagreed in government, is chosen by Starmer to replace her. 7 October – Foreign Secretary David Lammy says that the UK's decision to relinquish sovereignty of the Chagos Islands does not mean it has changed its position on the sovereignty of other overseas territories, such as the Falkland Islands and Gibraltar. Chagossians opposed to the government's decision to hand sovereignty of the Chagos Islands to Mauritius protest outside Parliament. Chagossians living in the UK complain that they were not consulted on the decision, with one saying "We don’t have a say, it’s as if we don’t count. We are just like their puppets. Where are our human rights?". 8 October – 2024 Conservative Party leadership election: Tom Tugendhat is eliminated from the leadership race in the latest round of voting, leaving three candidates to go forward to the next round.
=== Chemistry === The chemistry of polonium is similar to that of tellurium, although it also shows some similarities to its neighbor bismuth due to its metallic character. Polonium dissolves readily in dilute acids but is only slightly soluble in alkalis. Polonium solutions are first colored in pink by the Po2+ ions, but then rapidly become yellow because alpha radiation from polonium ionizes the solvent and converts Po2+ into Po4+. As polonium also emits alpha-particles after disintegration, this process is accompanied by bubbling and emission of heat and light by glassware due to the absorbed alpha particles; as a result, polonium solutions are volatile and will evaporate within days unless sealed. At pH about 1, polonium ions are readily hydrolyzed and complexed by acids such as oxalic acid, citric acid, and tartaric acid.

In December 2018, CIJA chief Stephen Rapp who formerly served as the US Ambassador for Global Criminal Justice, stated that war-crimes committed by the Syrian regime constituted a "solid kind of evidence that we haven't really had since Nuremberg, when the Nazis were prosecuted." The proofs of documented crimes included a vast array of sources, ranging from 2 million video footages to the documents seized from the Baathist regional committees and command crisis centres. Rapp asserted that despite Russian objections in the UN Security Council, the evidences are sufficient for an international arrest warrant.

=== Later medieval works === The Fisher King is a character in Chrétien's Perceval (1180) which is the first of a series of stories and texts on the subject of Perceval and the Grail. In his work, Chrétien did not say how the quester was related to this Fisher King. In later romances it is made clear, in the end, that he is the uncle of the quester. According to Eugène Vinaver, Chretien's work features two Maimed Kings. Parzival was written in 1210 by Wolfram von Eschenbach, thirty years after Perceval. Although a different work, it is strikingly similar to Perceval. The story revolves around the Grail Quest and once again the main character is Perceval (as Parzival). As in Perceval, the story has its protagonist fail to ask the healing question, which results in him Questing for years. However, Parzival differs from Perceval in three major ways. Firstly, the Fisher King is no longer nameless and is called Anfortas. Secondly, Eschenbach thoroughly describes the nature of the wound; it is a punishment for wooing a woman who is not meant for him (every Grail keeper is to marry the woman the Grail determines for him), and it causes him immense pain. Lastly, Parzival comes back to cure the Fisher King. Parzival, unlike its predecessor Perceval, has a definitive ending. The quester is the son of Herzeloyde, who is the sister of Anfortas.
=== Bone Morphogenetic Protein 1-like proteinases === In a study in 1996, Greenspan's lab showed that Bone Morphogenetic Protein 1 (BMP-1) is a protease responsible for the biosynthetic processing of the precursor protein procollagen I into type I collagen, the most abundant protein in the bodies of vertebrates. The lab also helped define the interactions of BMP1 with morphogens involved in dorsoventral patterning in early embryogenesis. They also were responsible, via various studies, for identifying and characterizing the other mammalian BMP1-like proteinases (BMP1-LPs), showing them to be a small family of proteases that activate and inactivate various growth factors, particularly a subset of the TGFβ superfamily; activate enzymes, such as lysyl oxidase, and biosynthetically process various extracellular structural macromolecules. In fact, the lab provided the preponderance of insights into biological roles of these extracellular proteases that are key to development and homeostasis and to ECM formation, and are important to formation of the cardiovascular system, wound healing, periodontal homeostasis and tooth development, and regulation of nodal Na+ channel clustering in formation of the nodes of Ranvier during neuronal development.
As long as the strategic American nuclear forces could overwhelm their Soviet counterparts, a Soviet pre-emptive strike could be averted. Moreover, the Soviet Union could not afford to build any reasonable counterforce, as the economic output of the United States was far larger than that of the Soviets, and they would be unable to achieve "nuclear parity". Soviet nuclear doctrine, however, did not match American nuclear doctrine. Soviet military planners assumed they could win a nuclear war. Therefore, they expected a large-scale nuclear exchange, followed by a "conventional war" which itself would involve heavy use of tactical nuclear weapons. American doctrine rather assumed that Soviet doctrine was similar, with the mutual in mutually assured destruction necessarily requiring that the other side see things in much the same way, rather than believing—as the Soviets did—that they could fight a large-scale, "combined nuclear and conventional" war. In accordance with their doctrine, the Soviet Union conducted large-scale military exercises to explore the possibility of defensive and offensive warfare during a nuclear war. The exercise, under the code name of "Snowball", involved the detonation of a nuclear bomb about twice as powerful as that which fell on Nagasaki and an army of approximately 45,000 soldiers on maneuvers through the hypocenter immediately after the blast. The exercise was conducted on September 14, 1954, under command of Marshal Georgy Zhukov to the north of Totskoye village in Orenburg Oblast, Russia.

It is a laboratory-made peptide that mimics growth hormone-releasing hormone. It prompts the pituitary gland to release growth hormone and has been studied mainly in adults with HIV-associated lipodystrophy.
Administered growth hormone supplies the hormone directly, while this peptide acts upstream by prompting the pituitary to release it. The indirect route preserves pulsatile secretion and some endogenous feedback, which changes the hormone and IGF-1 profile observed after treatment.
Reductions in visceral adipose tissue appear consistently in randomized trials of the approved population. Effects on peripheral fat, cardiovascular outcomes, and use outside that population are less well established.
It is a synthetic analog of growth hormone-releasing hormone, a hypothalamic peptide. It functions as a growth hormone secretagogue acting at pituitary receptors. The classification separates it from direct growth hormone products.
