Metabolic interest in this compound centers on fat distribution rather than on hormone levels alone. Imaging trials in adults with excess abdominal fat report reductions in visceral adipose tissue, while subcutaneous depots change comparatively little. Growth hormone and IGF-1 are presumed to carry the effect, but the separate contribution of each is not firmly established. Whether these changes persist after treatment stops, and whether they alter longer-term health outcomes, remain open questions that published work does not answer consistently.
Tesamorelin is a synthetic peptide of forty-four amino acids whose sequence reproduces human growth hormone-releasing hormone. Its distinguishing feature sits at the amino terminus, where a trans-3-hexenoyl group replaces the free amine. That acylation slows cleavage by dipeptidyl peptidase IV, an enzyme that otherwise removes the first two residues and inactivates the natural hormone quickly. The modified peptide therefore persists longer in circulation while keeping the same receptor target. It is handled as a lyophilized solid and dissolved shortly before use.
Signaling begins at the GHRH receptor, a class B G protein-coupled receptor displayed on somatotroph cells of the anterior pituitary. Receptor occupancy activates Gs proteins, which raise adenylyl cyclase activity and intracellular cyclic AMP, in turn driving protein kinase A dependent pathways. The downstream output is synthesis and pulsatile secretion of growth hormone into the bloodstream. Hepatic tissue and peripheral sites respond by increasing insulin-like growth factor 1 production. Somatostatin and IGF-1 itself supply negative feedback that caps the size and duration of each secretory burst.
Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.
Tesamorelin is a synthetic peptide of 44 amino acids that reproduces the sequence of human growth hormone-releasing hormone (GHRH) and carries a trans-3-hexenoyl group on its N-terminal tyrosine. That small fatty-acid modification blocks cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GHRH in plasma. The result is a molecule with a longer circulating half-life than the natural hormone while retaining the same receptor target. It is supplied as a lyophilized powder for reconstitution and belongs to the broader class of GHRH analogs studied for effects on pituitary growth hormone secretion.
Development work on the compound, originally designated TH9507, focused on conditions in which reduced growth hormone signaling is thought to contribute to altered body composition. The United States Food and Drug Administration approved it in 2010 for the treatment of excess visceral abdominal fat in adults with human immunodeficiency virus infection and lipodystrophy. Later research examined other populations, including adults with mild cognitive impairment, where a large trial did not meet its primary endpoints. This mixed record illustrates how a single mechanism can produce clear effects in one setting and inconclusive results in another.

It's possible that hackers have gotten into administrative computer systems of utility companies, but says those aren't linked to the equipment controlling the grid, at least not in developed countries. [Schmidt] has never heard that the grid itself has been hacked. In June 2019, Russia said that its electrical grid has been under cyber-attack by the United States. The New York Times reported that American hackers from the United States Cyber Command planted malware potentially capable of disrupting the Russian electrical grid.

== Detection of phase transitions == The basic principle underlying this technique is that when the sample undergoes a physical transformation such as phase transitions, more or less heat will need to flow to it than the reference to maintain both at the same temperature. Whether less or more heat must flow to the sample depends on whether the process is exothermic or endothermic. For example, as a solid sample melts to a liquid, it will require more heat flowing to the sample to increase its temperature at the same rate as the reference. This is due to the absorption of heat by the sample as it undergoes the endothermic phase transition from solid to liquid. Likewise, as the sample undergoes exothermic processes (such as crystallization) less heat is required to raise the sample temperature. By observing the difference in heat flow between the sample and reference, differential scanning calorimeters are able to measure the amount of heat absorbed or released during such transitions. DSC may also be used to observe more subtle physical changes, such as glass transitions. It is widely used in industrial settings as a quality control instrument due to its applicability in evaluating sample purity and for studying polymer curing.

Protein phosphorylation is a reversible post-translational modification of proteins in which an amino acid residue is phosphorylated by a protein kinase by the addition of a covalently bound phosphate group. Phosphorylation alters the structural conformation of a protein, causing it to become activated, deactivated, or otherwise modifying its function. Approximately 13,000 human proteins have sites that are phosphorylated. The reverse reaction of phosphorylation is called dephosphorylation, and is catalyzed by protein phosphatases. Protein kinases and phosphatases work independently and in a balance to regulate the function of proteins. The amino acids most commonly phosphorylated are serine, threonine, tyrosine, and histidine. These phosphorylations play important and well-characterized roles in signaling pathways and metabolism. However, other amino acids can also be phosphorylated post-translationally, including arginine, lysine, aspartic acid, glutamic acid and cysteine, and these phosphorylated amino acids have been identified to be present in human cell extracts and fixed human cells using a combination of antibody-based analysis (for pHis) and mass spectrometry (for all other amino acids). Protein phosphorylation was first reported in 1906 by Phoebus Levene at the Rockefeller Institute for Medical Research with the discovery of phosphorylated vitellin. However, it was nearly 50 years until the enzymatic phosphorylation of proteins by protein kinases was discovered.

According to neo-Darwinian evolution, such mutations should rarely exist as these sequences are functionless and positive selection would not be able to operate. The McDonald–Kreitman test has been used to study selection over long periods of evolutionary time. This is a statistical test that compares polymorphism in neutral and functional sites and estimates what fraction of substitutions have been acted on by positive selection. The test often uses synonymous substitutions in protein coding genes as the neutral component; however, synonymous mutations have been shown to be under purifying selection in many instances.

Hence, pseudoprolines fulfill two functions simultaneously: they serve (1) as temporary side-chain protection for Ser, Thr, and Cys and (2) as solubilizing building blocks to increase solvation and coupling rates during peptide synthesis and in subsequent chain assembly. Pseudoprolines are obtained by reacting the free amino acids with aldehydes or ketone. The coupling of amino acid derivatives to a growing peptide chain containing N-terminal pseudoproline generally results in low yields because of the sterically hindered nature of the oxazolidine (thiazolidine) ring system and the decreased nucleophilicity of the nitrogen atom. Consequently, the preformation of suitably protected dipeptide derivatives of the type FMOC-Xaa1-Oxa/THz-OH is preferable for use in peptide synthesis. Two conceptually different approaches are feasible for preparing oxazolidine- and thiazolidine-ring-containing dipeptide derivatives: (1) the in situ acylation of Ser- or Thr-derived oxazolidines or Cys-derived thiazolidines using acid fluorides or N-carboxyanhydrides (NCA); and (2) the direct insertion of the oxazolidine systems into dipeptides (post-insertion) containing C-terminal Ser or Thr. The method of choice strongly depends on the nature of the pseudoproline as welI as on the substituents at C2 of the cyclic system.
Jung saw Freud's theory of the unconscious as incomplete, unnecessarily negative, and inelastic. According to Jung, Freud conceived the unconscious solely as a repository of repressed emotions and desires. Jung's observations overlap to an extent with Freud's model of the unconscious, what Jung called the "personal unconscious", but his hypothesis is more about a process than a static model, and he also proposed the existence of a second, overarching form of the unconscious beyond the personal, that he named the psychoid—a term borrowed from neo-vitalist philosopher and embryologist Hans Driesch (1867–1941)—but with a somewhat altered meaning. The collective unconscious is not so much a 'geographical location', but a deduction from the alleged ubiquity of archetypes over space and time. Consequently, their personal and professional relationship fractured—each stating the other could not admit he could be wrong. In November 1912, Jung and Freud met in Munich for a meeting among prominent colleagues to discuss psychoanalytical journals. At a talk about a new psychoanalytic essay on Amenhotep IV, Jung expressed his views on how it related to actual conflicts in the psychoanalytic movement. While Jung spoke, Freud suddenly fainted, and Jung carried him to a couch. In early January 1913, Freud wrote to Jung, proposing that "we abandon our private relationship entirely. I will lose nothing, because I have long been joined to you only by the thin thread of the further development of past disappointments". Jung agreed to comply with this arrangement.

=== Thyroglobulin antibodies === In the clinical laboratory, thyroglobulin testing can be complicated by the presence of anti-thyroglobulin antibodies (ATAs, alternatively referred to as TgAb). Anti-thyroglobulin antibodies are present in 1 in 10 normal individuals, and a greater percentage of patients with thyroid carcinoma. The presence of these antibodies can result in falsely low (or rarely falsely high) levels of reported thyroglobulin, a problem that can be somewhat circumvented by concomitant testing for the presence of ATAs. The ideal strategy for a clinician's interpretation and management of patient care in the event of confounding detection of ATAs is testing to follow serial quantitative measurements (rather than a single laboratory measurement). ATAs are often found in patients with Hashimoto's thyroiditis or Graves' disease. Their presence is of limited use in the diagnosis of these diseases, since they may also be present in healthy euthyroid individuals. ATAs are also found in patients with Hashimoto's encephalopathy, a neuroendocrine disorder related to—but not caused by—Hashimoto's thyroiditis.
==== Glucose absorption ==== In 1960, the biochemist Robert K. Crane revealed his discovery of the sodium-glucose cotransport as the mechanism for intestinal glucose absorption. This was the very first proposal of a coupling between the fluxes of an ion and a substrate that has been seen as sparking a revolution in biology. This discovery, however, would not have been possible if it were not for the discovery of the molecule glucose's structure and chemical makeup. These discoveries are largely attributed to the German chemist Emil Fischer who received the Nobel Prize in chemistry nearly 60 years earlier.
=== Other === Citalopram may be used off-label to treat anxiety, and dysthymia, premenstrual dysphoric disorder, body dysmorphic disorder, and obsessive–compulsive disorder. It appears to be as effective as fluvoxamine and paroxetine in obsessive–compulsive disorder. Some data suggest the effectiveness of intravenous infusion of citalopram in resistant OCD. Citalopram is well tolerated and as effective as moclobemide in social anxiety disorder. There are studies suggesting that citalopram can be useful in reducing aggressive and impulsive behavior. It appears to be superior to placebo for behavioural disturbances associated with dementia. It has also been used successfully for hypersexuality in early Alzheimer's disease. A meta-analysis, including studies with fluoxetine, paroxetine, sertraline, escitalopram, and citalopram versus placebo, showed SSRIs to be effective in reducing symptoms of premenstrual syndrome, whether taken continuously or just in the luteal phase. For alcoholism, citalopram has produced a modest reduction alcohol intake and increase in drink-free days in studies of alcoholics, possibly by decreasing desire or reducing the reward. While on its own citalopram is less effective than amitriptyline in the prevention of migraines, in refractory cases, combination therapy may be more effective. Citalopram and other SSRIs can be used to treat hot flashes.
== Acquisition == In June 2013, Amgen offered to buy the shares of Onyx Pharmaceuticals for US$120 per share, sending the shares up by around 30% after the news was announced. Onyx announced on June 30, 2013, that it had rejected the unsolicited proffer from Amgen and its board of directors had subsequently authorized their financial advisers to contact other potential acquisition suitors. However after a series of efforts to secure a higher per-share price from other potential buyers repeatedly failed to materialize, the board reversed course and agreed to Amgen's original terms; the acquisition was formally announced on August 25, 2013.

The amino acid sequence matches human growth hormone-releasing hormone, but the amino terminus carries a trans-3-hexenoyl group instead of a free amine. That single structural change chiefly affects enzymatic stability rather than receptor selectivity.
Growth hormone released from the pituitary stimulates IGF-1 production in the liver and other tissues, so circulating IGF-1 generally rises during exposure. The size of the rise varies between individuals and depends on baseline hormonal status and other concurrent factors.
Reported imaging studies focus on visceral adipose tissue, where reductions are more consistently observed across trials. Subcutaneous depots show smaller and less reproducible changes, so the two compartments should not be treated as equivalent.
It is a synthetic peptide built from 44 amino acids arranged in the same order as human growth hormone-releasing hormone. A short fatty-acid chain, described as a trans-3-hexenoyl group, is attached to the first amino acid. The finished molecule is formulated as a sterile powder that is dissolved before use.
